Phenotypic and Genotypic differences in the expression of virulence factors in antimicrobial resistance of Enterococcus faecalis clinical strains

چکیده
There is no information on investigations comparing phenotypic and genotypic differences in MDR and MDS E. faecalis strains in literature. The study included 70 clinical E. faecalis isolates from human origin. None of the isolates tested demonstrated any in vitro sensitive to vancomycin, with MIC values of all the strains for vancomycin of ≥23 μg/mL. High-level resistances to ampicillin and streptomycin were detected by the disc diffusion method in 77% and 80% of the isolates, respectively. The MDR E. faecalis strains which were investigated produced significantly amounts of extracellular material binding Congo red and gelatinase quantity in culture supernatants when compared with MDS strains. The capacity to produce lipase and DNase activity were not found in any of the isolates investigated. Cytolysin activity was detected in 70% of MDR E. faecalis strains, where in only 20% of MDS E. faecalis. MDR strains adhered to polystyrene to a more extent than MDS strains. The presence of virulence genes (esp, cylA, gelE and EF3314) were investigated by PCR. MDR E. faecalis strains showed more detection of these genes than MDS E. faecalis strains. The presence of these genes did not correlate completely with their phenotypic expression. In conclusion, the MDR E. faecalis strains have impaired virulence when compared to MDS strains.

Molecular detection of virulence factors and antibiotic resistance pattern in clinical Enterococcus faecalis strains in Sardinia.
In this study, the antibiotic resistance pattern and the presence of genes encoding several virulence factors in 91 Enterococcus faecalis strains isolated from different human clinical sources in Sardinia were investigated. Genotypic determination of virulence genes (gelE, esp, agg, ace, cylA,B,M,L(L),L(S), efaA, fsrB) was carried out by PCR. The production of gelatinase and haemolytic activity...
متن کاملEvaluation of Virulence Factors and Plasmid-Related Transmissibility among Different Isolates of Enterococci
The incidence of virulence factors among 114 Enterococcus faecalis and 35 Enterococcus faecium strains from different clinical specimens were compared with those isolated from control groups. A few of the isolates expressed two or more of the following traits simultaneously: hemolysin, aggregation substance, gelatinase, DNase, hemagglutinin and antibiotic resistance. The frequencies of hemolysi...
متن کاملAntibiogram analysis and tracking of the virulence-related genes in Enterococcus faecalis isolates
Background and Aim: Enterococcus species are opportunistic pathogens and their pathogenicity seems to be related to the presence of a number of pathogenicity genes. Since donkey’s milk is a new non-allergenic source of nutrition, this study was performed to assess the antibiogram and detection of pathogenicity genes in some Enterococcus faecalis isolates from donkey’s milk. Materials and Method...
متن کاملHigh Incidence of Virulence Factors Among Clinical Enterococcus faecalis Isolates in Southwestern Iran
BACKGROUND Over the past two decades, enterococci have emerged as an important agent responsible for hospital acquired infection. Several virulence factors contribute to the adherence, colonization, evasion of the host immune response, and pathogenicity and severity of the infection. Enterococcus faecalis is the most common and virulent species causing infections in hospitalized patients. The a...
متن کاملEvaluation of Phenotypic and Genotypic Expression of Efflux Pumpin Clinical Isolates of Acinetobacter baumannii Resistant to Ciprofloxacin and Gentamicin
Introduction: Acinetobacter baumannii is an important bacteria because of high ability to obtain antibiotic resistance genes and creation of multi-drug resistant strains (MDR). Today, the active efflux pumps has been suggested as one of the most important mechanisms of intrinsic and acquired resistance of antibiotics in bacteria. The aim of this study was Evaluation of phenotypic and genotypic ...
